Challenge Mode can only be activated once you finished the E4 and the Champion. And yes, you do keep your save file.

So i have a question and wasnt sure where to put this. I got the Landeros in the dream radar and got the reveal glass. if i trade him to my black version and restart the game can i get the reveal glass again or is he stuck in the incarnate form for good?

You'd have to transfer another Landorus from the Dream Radar if you wanted the Reveal Glass on the new save.
