Byron. His Bastiodon killed my Torterra many times last playthrough (when I still only used on Pokemon at a time). Plus... he is the Steel-type Gym leader, after all. (okay, bad pun, I know.)

Fantina's Mismagius and Candice's Froslass in Platinum are some of the most annoying for sure. I don't know who I've lost to the most, though. (I always reset until I win...)

Actually the only gym battle that I lost was Fantina. The other gyms I all beat on my first try because I was training a team. But in Pearl when I only trained my starter because I was new to Pokemon Gardeina took FOREVER (I started with Piplup).

I never lost to Fantina, but I started to play with Piplup at the first time, Gardenia's Roserade owned my Prinplup, which was my strongest pokemon, btw.

None of the Gym Leaders "killed" me, though Candice came close when I soloed Diamond with Torterra one time.
Overall though, I'd say that Maylene's the hardest for me. I find her Lucario in Platinum to be really troublesome.

No one killed me as I beat the game with a level 70 luxray and a level 40 vaporeon. The rest of my team was just HM slaves.

I lost once to Fantina. I find Maylene to be the second most challenging.

I never lost to any of the leaders, but I was very close to losing against Gardenia in Platinum. She defeated my Prinplup without any problems, and IIRC my Luxio survived a hit from one of her Pokemon with exactly one HP left. I also found Volkner in Platinum very hard to defeat.

Gardenia defeated me a lot more than the others (Volkner being the closest), since I mostly battled with the Piplup line.

None of the gym leaders seemed difficult to me, but wake, I mean crasher wake, got me a couple times in platinum.

More than half of the people voted Fantina including me. No trouble in Pearl for me with her. She was a real pain in Platinum. If you can whip out a higher leveled Mismagius, and get a dark type around level 25 in Platinum 34 in DP, you're all set for her.
